Abstract

In this study, the influence of amphetamines on GABA (gamma amino butyric acid) was studied in the rat brain (striatum, cortex and hippocampus). According to literature, an increase in GABA is expected after 4 hours, whereas normalization occurs within 24 hours. Rats were therefore injected intraperitoneally with 30mg/kg amphetamine and necropsied after 4 and 24h. Salt solution was injected as a negative control in control animals. Brain samples were derivated by means of OPA/ß-mercaptoethanol and iodoacetamide, followed by HPLC and electrochemical analysis. First, there was made a linearity curve with the standards of GABA. And when the samples were measured, the concentration can be calculated with the linearity curve. For this results there can be concluded that amphetamines have no effect on GABA concentrations in the different brain levels. But because there were too less samples, the conclusion is not very reliable. Because of the less results, statistical analysis could not be done. In dit onderzoek werd de invloed van amfetamines op GABA (gamma amino boterzuur) bestudeerd in de hersenen van de rat (striatum, cortex en hippocampus). Volgens de literatuur wordt een stijging van GABA verwacht na 4 uur, terwijl normalisatie voorkomt na 24 uur. Hiervoor werden de ratten intraperitonaal geïnjecteerd met 30mg/kg amfetamine en gedissecteerd na 4 en 24 uur. De zoutoplossing werd geïnjecteerd als negatieve controle in controle dieren. Stalen van de hersenen werden gederivatiseerd met OPA/ß-mercaptoethanol en joodacetamide, gevolgd door HPLC en electrochemische detectie. Allereerst werd er een lineariteitscurve gemaakt met de standaarden van GABA. En wanneer de stalen gemeten werden, kon de concentratie berekend worden door middel van de lineariteitscirve. Voor deze resultaten kan er geconcludeerd worden dat amfetamines geen effect hebben op de GABA concentraties in de verschillende hersendelen. Abstract

Cryopreservation is an efficient way of preserving transgenic mouse strains. Even if the strain is still in use, it is a good idea to preserve germ line cells or embryos. This technique can back-up genetic drift and disasters in the housing facilities. The original strain can be easily revitalized and be placed in experiment again after such incidence. Cryopreservation is also less costly than keeping a strain of transgenic mice alive over a longer period of time. The aim of this thesis is to make a comparison between the two common cryopreservation methods: rapid freezing of mouse spermatozoa and vitrification of 2-cell mouse embryos. Literature shows that rapid freezing of spermatozoa has a higher probability of causing intra- and extracellular ice crystals. Vitrification of embryos doesn’t have the problem of ice crystal formation as the samples are cooled immediately to -180°C. In this condition, water molecules transition into a glasslike state without the formation of ice crystals, which are the main cause of physical damage to the cells. Consequently vitrification is the safest method to prevent this physical damage. Both vitrification and rapid freezing use high concentrations of cryoprotective agents (CPA) to prevent damage by ice crystals on cellular content. These high concentrations of CPA are potentially toxic to the samples at room temperature, hence it is necessary to vitrify or freeze the cells as fast as possible. After vitrification, almost all embryos are viable. Half of the 2-cell stage embryos that are implanted in a pseudo-pregnant female, develop into pups. Using the technique of rapid freezing, only half of spermatozoa are viable after thawing. The fertilization rate of thawed sperm used in an in vitro fertilization experiment is approximately 65%. On the other hand, with one sperm freezing, hundreds of zygotes can be fertilized. The most important advantage of conserving embryos is the fact that they are diploid while spermatozoa are haploid. All genes are preserved with the embryo while only paternal genes are preserved in spermatozoa. Pups deriving from vitrified embryos are preserving the whole genome of the original strain, while offspring of frozen spermatozoa will be heterozygous for the intended mutation unless oocytes of the same strain are preserved. In order to obtain homozygous offspring, intercrossing can be perfomed. The costs of rapid freezing of spermatozoa are cheaper than embryo-vitrification, but taking the price of revitalization into account, rapid freezing of spermatozoa and vitrification of 2-cell embryos cost almost the same. Keeping the 3R (reduction, refinement and replacement) idea in mind, the number of mice used for an experiment are also an important criteria. To obtain a good number of embryos 20-50 female mice are needed. On the other hand, from one fertile male several samples can be stored. To make a final decision on whether to use rapid freezing of spermatozoa or vitrification of 2-cell embryos, all the above factors should be taken into account. Of notice, it is designated to select males and females for cryopreservation during the active breeding programs. For both techniques there is no need of very high-tech equipment, nevertheless both ask a certain level of technical skills. To cryopreserve the samples with the aim to store them for future investigation a good data banking and safety system on the cryo-tanks is needed.

Abstract

With the surge of new distribution technologies of non-linear television, for a large part brought about by Netflix, traditional practices concerning television viewing and television industry lore have been challenged. More often, people are choosing to figuratively cut the cord and cancel their linear television subscriptions. These trends are causing broadcasting networks anxieties. The altering behavior of the audience has forced networks and broadcasting organizations to revise their established practices and to innovate. The television industry has been marked by increasing commercialization, fragmentation and platformization. Ever since the late 1990s, Public Service Broadcasters have experienced threats, back then because of the rise of commercial channels and networks. During the last decade, public service broadcasters around Europe have experienced new threats, and their existence and value in society was questioned. Moreover, international conglomerates have entered the local media markets and threaten to overshadow domestic companies. Local commercial networks answered these threats by establishing their own non-linear distribution portals, often subscription-based and including a ‘local Netflix’. For public service broadcasters, it is more difficult to find a sustainable balance between commercial goals and innovation, as they have to answer to more restrictions in accordance with the government. In August 2022, Flemish Public Service Broadcaster VRT rebranded their non-linear internet distribution portal, VRT NU, to VRT MAX after it became apparent that it did not serve the needs of a large demographic, namely those between the ages of 15 and 35, thus including Generation Z. To breach this lacuna in audience, VRT MAX targets this group specifically, thereby consolidating its position in society and as Public Service Media. This thesis first looked into the various developments that took place in the television industry, particularly after the arrival of Netflix. Then, the research focused on evolution within the Flemish ecosystem. In the final part, a case study evaluated in which ways VRT MAX as a rebranded portal has contributed to the consolidation of VRT as Public Service Media in the Flemish media ecosystem, which has been disrupted by commercialization, fragmentation and platformization. The case study will follow the strategies public service media across Europe have taken in reaction to international subscription-based video-ondemand services, a typology that stems for Philip Napoli but which was adapted by D’Arma, Raats, and Steemers. From this case study was concluded that VRT MAX is still very new in the media ecosystem, and can contribute to the consolidation of the position of VRT drawing on the strategies of differentiation, mimicry and collaboration.
